Objective: The present study aimed to investigate the ability of hesperidinto suppress the migration and invasion of A549 cells, and to investigate the role of the SDF- 1/CXCR-4 cascade in this suppression.

Methods: We performed a Transwell migration assay to measure the migratory capability of A549 cells treated with 0.5% DMSO, SDF- 1α, AMD3100 or hesperidin. The SDF- 1 level in the culture medium was determined by an enzyme-linked immunosorbent assay (ELISA) to detect whether different concentrations of hesperidin affected SDF- 1 secretion. A wound-healing assay was performed to determine the effects of different concentrations of hesperidin on the migration inhibition of A549, H460 and H1975 cells. Additionally, the effect of various hesperidin concentrations on the rate of A549 cell invasion and migration was examined with and without Matrigel in Transwell assays, respectively. Western blot analysis was used to evaluate the protein levels of CXCR-4, MMP-9, CK- 19, Vimentin, p65, p-p65, p-IκB, IκB, p-Akt and Akt. RT-qPCR was used to detect the mRNA levels of CXCR-4, MMP-9, CK- 19, Vimentin, p65, IκB, SDF- 1 and Akt.

Results: The Transwell migration assay indicated that SDF- 1α promoted A549 cell migration, while AMD3100 and hesperidin significantly inhibited the migratory capability. The wound-healing assay demonstrated that hesperidin treatment significantly reduced the rate of wound closure compared with the control group in a dose-dependent manner. Similarly, the migration and invasive abilities of A549 cells, H460 and H1975 cells treated with hesperidin were significantly decreased compared with the control group. The ELISA data suggested that hesperidin attenuated the secretion of SDF- 1 from A549 cells in a dose-dependent manner. Furthermore, western blot analysis indicated that SDF- 1α treatment significantly increased the levels of CXCR-4, p-p65, p-IκB and p-Akt in A549 cells. In contrast, AMD3100 or hesperidin reversed the effect induced by SDF- 1α through decreasing the expression of CXCR-4. Subsequent RT-qPCR and western blot analyses also confirmed that hesperidin had a significant effect on the expression of EMT-related proteins,including MMP-9, CK- 19 and Vimentin, in A549 cells.

Conclusion: In summary, we demonstrated that hesperidin inhibited the migratory and invasive capabilities of A549 human non-small cell lung cancer cells by the mediation of the SDF- 1/CXCR-4 signaling cascade, thus providing the foundation for the development of hesperidin as a safer and more effective anticancer drug for non-small cell lung cancer.

Introduction

Globally, lung cancer has the highest morbidity and mortality rates of all types of cancer, with 1.5 million patients newly diagnosed annually[1] . Approximately 85% of lung cancer cases are non-small cell lung cancer (NSCLC)[2] . The first-line treatment for NSCLC is surgical resection combined with radiotherapy and chemotherapy[1, 3] . Despite significant improvements in the efficacy of chemotherapy drugs and the availability of molecularly targeted therapy drugs in clinical practice in the past decade, the 5-year survival rate of patients with NSCLC is still low[1, 3] . Tumor metastasis is a critical factor in the low survival rate for patients with NSCLC, which is associated with a variety of factors, including angiogenesis[4, 5], epithelial-mesenchymal transition (EMT)[6-8], and the migration[9] and invasion[10, 11] of tumor cells. The activation of the CXCR4/SDF1 axis plays an important role in the process of NSCLC metastasis[12-14] .

Despite the development of small-molecule drugs with greater specificity and efficacy against tumors, the side effects of chemotherapy drugs are severe, limiting their clinical application. Natural plant extracts from a wide range of sources may have fewer side effects and higher tolerability and have provided novel insights into cancer treatment in recent years[15] . Hesperidin is a natural flavonoid glycoside compound, occurring in orange peel and other citrus species, that is widely used in Chinese herbal medicine[16] . It was previously demonstrated to induce anticancer activity through the promotion of apoptosis[17-21] . Furthermore, hesperidin affected the expression of MMP family members and EMT-related proteins to inhibit cell invasion, resulting in anti-cancer activity[22-24] . In vitro and in vivo studies have demonstrated that hesperidin enhances antioxidant activity, thus inhibiting tumor development[25, 26]. Our previous studies have identified that hesperidin promotes the apoptosis of A549 NSCLC cells by regulating the proteins in the mitochondrial apoptosis pathway and modulating the expression of cell cycle-related molecules, without any negative effects on BEAS-2B human normal lung epithelial cells[21] . However, data regarding the effect of hesperidin on NSCLC cells is incomplete. Hence, we investigated the SDF- 1/CXCR-4 signaling pathway and EMT process to understand the underlying mechanisms of the effect of hesperidin. The reported results may provide the foundation for the development of hesperidin as a safer and more effective anticancer drug for NSCLC.

1. Materials and methods
1.1 Materials

A549, H460 and H1975 NSCLC cells were purchased from the Cell Bank of the Chinese Academy of Sciences (Shanghai, China). RPMI 1640 medium and trypsin were purchased from Hyclone (GE Healthcare Life Sciences, Logan, UT, USA). Fetal bovine serum (FBS) was purchased from Gibco (Thermo Fisher Scientific, Inc., Waltham, MA, USA). Hesperidin (Catalog No. SC-205711A) and AMD3100 (Catalog No. CAS 155148-31-5) was purchased from Santa Cruz Biotechnology, Inc. (Dallas, TX, USA). Rabbit anti-human β-actin (Catalog No. ab8226), CXCR-4 (Catalog No. ab124824) and MMP-9 (Catalog No. ab73734) primary antibodies, and goat anti-rabbit HRP-IgG (Catalog No. ab6721) were purchased from Abcam (Cambridge, UK). Rabbit anti-human CK- 19 and Vimentin primary antibodies, and recombinant SDF- 1α were purchased from Gene Tech Co., Ltd (Catalog No. 300-28A, Shanghai, China). Rabbit anti-human p65 (Catalog No. 10745- 1-AP) and p-p65 (Catalog No. BS1570- 100) primary antibodies were purchased epigenetic drug target from Wuhan Sanying Biotechnology Co., Ltd. (Wuhan, China). Rabbit anti-human Akt (Catalog No. 9272), p-Akt (Catalog No. 4691), IκB (Catalog No. 4812) and p-IκB (Catalog No. 2859) primary antibodies were purchased from Cell Signaling Technology, Inc. (Danvers, MA, USA). Transwell migration chambers and Matrigel were provided by BD Biosciences (Franklin Lakes, NJ, USA). An SDF- 1 ELISA Kit was supplied by ImmunoWay Biotechnology Co. (Catalog No. KE1216, Plano, TX, USA). All primers were synthesized by Sangon Biotech Co., Ltd. (Shanghai, China). TRIzol was purchased from Life Technologies (Thermo Fisher Scientific, Inc.). Reverse transcription and SYBR Green qPCR kits were purchased from Takara Bio, Inc. (Otsu, Japan).

1.2 Cell culture and groups

All cells were cultured in RPMI 1640 medium containing 10% FBS and 1% streptomycin in a thermostatic incubator at 37°C with 5% CO2. All experiments were
repeated three times.

1.3 Migration assay

1.3.1 In order to detect the appropriate concentration of SDF- 1α for the migration assays, A549 cells were cultured in 24-well plates at a density of 1×105 cells/well for 24 h and treated with different concentrations of SDF- 1α (0, 40, 60, 80 and 100 ng/mL) and 100 ng/mL SDF- 1α with 30 μg/mL AMD3100 as a positive control. All cells were harvested after 48 h, resuspended in 200 μL serum-free medium, and seeded in the upper Transwell migration chamber, with 6% FBS in the lower chamber.Secondly, 100 ng/mL SDF- 1α was selected to investigate the effect of SDF- 1α on the transmembrane migration of A549 cells. Five groups were used for this assay, as follows: 0.5% DMSO, SDF- 1α, SDF- 1α & hesperidin (25 μg/mL), SDF- 1α & hesperidin (50 μg/mL), SDF- 1α & AMD3100 (30 μg/mL). The Transwell migration assay was performed as specified above. Following a 4-h incubation, cells crossing the membrane into lower chamber were stained with 0.1% crystal violet at 37°C for 15 min, and photographed with an inverted microscope (Olympus Corporation, Tokyo,Japan).

1.3.2 To investigate the appropriate concentration of FBS and SDF- 1α for the Chemotaxis assay, A549 cells were seeded into the upper Transwell chamber and added different concentrations of FBS (0, 2, 4, 6, 8 and 10%) or various concentrations of SDF- 1α (30, 60, 120, 240, 480 and 960 ng/mL) serum-free medium in lower chamber. Following a 4-h incubation, cells crossing the membrane into lower chamber were stained with 0.1% crystal violet at 37°C for 15 min, and photographed with an inverted microscope (Olympus Corporation, Tokyo, Japan).Moreover, cells were pretreated with different concentrations of hesperidin (25, 37.5, 50 and 62.5 μg/mL) for 72 h, then seeded into the upper chamber with 600 μL serum-free medium containing 500 ng/mL SDF- 1α or 6% FBS medium in the lower chamber and incubated for 4 h. The cells were stained and photographed as described above.

1.4 Wound-healing assay

A549, H460 and H1975 cells were seeded in 6-well plates at a density of 5×105 cells/well and incubated for 24 h to form a confluent monolayer, then scratched with a 1 mL pipette tip across the center of the plate. The medium was replaced with medium containing different concentrations of hesperidin (25, 37.5, 50 and 62.5 μg/mL) every 12 h. The results were photographed under an inverted microscope at 0, 24, 48 and 72 h.

1.5 Invasion assay

A549 cells were seeded in 24-well plates at a density of 1×105 cells/well and treated with hesperidin (25, 37.5, 50 and 62.5 μg/mL) for 48 h. The cells were then transferred to Transwell chambers pre-coated with 100 μL Matrigel (500 ng/mL) and incubated at 37°C for 2 h; the lower chamber was supplemented with 600 μL medium containing 6% FBS and 500 ng/mL SDF- 1α. After 4 h, cells were stained and observed as described above.

1.6 ELISA assay

A549 cells were seeded in 24-well plates at a density of 1×105 cells/well and incubated for 24 h. Medium containing hesperidin (25, 37.5, 50 and 62.5 μg/mL) was changed every 12 h for 5 days. Cells were collected and centrifuged at 10,000 x g for 1 min prior to analysis of the secreted SDF- 1 level in the supernatant using the ELISA kit.

1.7 RT-qPCR assay

A549 cells were seeded into 6-well plates at a density of 5×105 cells/well for 24 h, then treated with different concentrations of hesperidin (25, 37.5, 50 and 62.5 μg/mL) for 72 h. Total RNA was extracted from the cells using TRIzol reagent according to the manufacturer’s instructions. The concentration of RNA was determined by measuring the optical density (OD) at 260 nm, and the purity of RNA was determined by measuring the 260/280nm ratio, with a ratio between 1.8 and 2.0 considered sufficiently pure. cDNA was synthesized according to the reverse transcription kit protocol. Reverse transcription was performed in a Mastercycler Gradient 5331 (Eppendorf, Hamburg, Germany) at 37°C for 15 min and 85°C for 5 sec, followed by storage at 4°C. qPCR was performed according to the manufacturer’s guidelines using the BIO-RAD q-PCR instrument (Bio-Rad Laboratories, Inc., Hercules, CA, USA), with thermocycling conditions as follows: 95°C for 15 sec; then 95°C for 5 sec and 62°C for 30 sec for 40 cycles.

1.8 Western blot

Total protein was collected from each group and quantified by a BCA assay. Protein samples were separated by 10% SDS-PAGE and transferred onto a PVDF membrane (0.25 μm). Bovine serum albumin was used for blocking for 2 h at room temperature. The corresponding antibodies (dilution, 1:2000) were incubated with the membrane overnight at 4°C, then washed 3 times in TBST and incubated with the appropriate HRP-conjugated secondary antibody for 2 h. The bound antibodies were visualized using an enhanced chemiluminescence (ECL) detection reagent (Bio-Rad Laboratories, Inc.; cat. no. 1705060) and evaluated by using Image Laboratory software (Bio-Rad Laboratories, Inc.).

1.9 Statistical analysis

All values are expressed as the mean ± standard deviation (SD). Statistical analysis was performed using SPSS for Windows, version 19.0 (IBM Corporation, Armonk, NY, USA). The differences between groups were analyzed by one-way ANOVA with Dunnett’s test for multiple group comparison. The significance threshold was set as P<0.05. 2. Results
2.1 Effect of SDF-1α and hesperidin on the migratory capability of A549 cells

As shown in figure 1, SDF- 1α treatment significantly increased the rate of transmembrane migration in A549 cells compared with the control group in a dose-dependent manner (P<0.05). However, treatment with a specific CXCR-4 inhibitor, AMD3100, could significantly reduce the number of cells that migrated (P<0.05, figure 1 A1, A2) compared with the 100 ng/mL SDF- 1α-treated group. In addition, as shown in Figure 1B1 and 1B2, SDF- 1α increased the rate of transmembrane migration in A549 cells compared with the control (P<0.05), whereas simultaneous treatment with 50 or 100 μg/mL hesperidin and 30 μg/mL of AMD3100 significantly inhibited SDF- 1α-induced cell migration compared with the 100 ng/mL SDF- 1α-treated group (P<0.05). 0.5% DMSO had no effect on the transmembrane migration of A549 cells compared with the control (P>0.05).

Values are expressed as the mean ± SD. *P<0.05 vs. control, #P<0.05 vs.SDF- 1α group. 2.2 Effect of SDF-1α and hesperidin on the SDF-1/CXCR-4 signaling cascade in A549 cells The protein and transcriptional levels of molecules associated with the SDF- 1/CXCR-4 signaling cascade are illustrated in figure 2. SDF- 1α treatment significantly upregulated the level of CXCR-4, p-p65 and P-Akt protein expression (P<0.05), and downregulated p-IκB (P<0.05) while no significant difference in protein expression was identified for p65, IκB or Akt compared with the control (P>0.05). 50 μg/mL hesperidin or 30 μg/mL AMD3100 treatment significantly inhibited the SDF- 1α-induced protein expression of p-p65, p-Akt and p-IκB, compared with the SDF- 1α-only group (P<0.05). 0.5% DMSO had no significant effect on protein expression compared with the control group (P>0.05). The relative transcriptional expression data, as determined by RT-qPCR, were consistent with the results of western blot analysis.

Statistical analysis was performed by ANOVA with Dunnett’s test. *P<0.05 vs. control, #P<0.05 vs. SDF- 1α group. 2.3 Effect of hesperidin on the expression of endogenous SDF-1 in A549 cells The results of the ELISA (figure 3, A) and RT-qPCR (figure 3, B) assays demonstrated that the expression of SDF- 1 in the 0.5% DMSO group was not significantly different compared with the control group, whereas SDF- 1 expression in the hesperidin-treated groups was significantly lower than in the control group (P<0.05). Statistical analysis was performed by ANOVA with Dunnett’s test. *P<0.05 vs. control. 2.4 Effect of hesperidin on the migration of A549, H460 and H1975 cells Results of the wound-healing assay are displayed in Figure 4. The 0.5% DMSO group showed no significant difference in wound-fusion rate compared with the control group in A549 cells (P>0.05). The wound-fusion rates among hesperidin groups were significantly lower than that in the control group (P<0.05), in a dose-dependent manner (figure 4A). The results in H460 (figure 4B) and H1975 (figure 4C) cells were consistent with A549 cells. Statistical analysis was performed by ANOVA with Dunnett’s test. *P<0.05. 2.5 Effect of hesperidin on the transmembrane migratory capability of A549 cells As shown in figure 5, the FBS-induced transmembrane migratory capability of A549 cells was increased in a concentration-dependent manner. There was no significant difference between the 0.5% DMSO and control groups when cultured with 6% FBS. The migratory efficiency in hesperidin-treated groups was markedly attenuated compared with the control group in a dose-dependent manner (P<0.05;figure 5, B1, B2). Statistical analysis was performed by ANOVA with Dunnett’s test. *P<0.05. 2.6 Effect of hesperidin on the chemotaxis ability of A549 cells The SDF- 1α-induced trans-membrane migratory capability of A549 cells was improved in a concentration-dependent manner (figure 6, A1, A2). There was no difference between the 0.5% DMSO and control groups with 500 ng/mL SDF- 1α in the lower chamber (P>0.05). The migration rate of A549 cells treated with the different concentrations of hesperidin was lower than that of the control (P<0.05,figure 6, B1,B2). There was no significant difference between the 0.5% DMSO and control groups when cultured with 6% FBS (figure 7, A1, A2), or 500 ng/mL SDF- 1α (figure 7, B1, B2) (P>0.05). The invasion rate in the hesperidin-treated groups was significantly decreased in a concentration-dependent manner compared with the control (P<0.05). Statistical analysis was performed by ANOVA with Dunnett’s test. *P<0.05 vs. control. 2.7 Effects of hesperidin on the SDF-1/CXCR-4 signaling cascade in A549 cells Our results indicated that the protein expression levels of CXCR-4, MMP-9 and Vimentin were significantly downregulated in hesperidin-treated groups compared tendon biology with the control. By contrast, the protein expression of CK- 19 was significantly upregulated compared with the control (P<0.05). 0.5% DMSO had no significant effect on the relative protein expressions compared with the control (P>0.05). The results on the transcriptional level, as determined by RT-qPCR, were similar to the results of western blot analysis.

Discussion Our results suggested that the inhibitory effect of hesperidin on the migration and invasion of A549 NSCLC cells was affected by the SDF- 1/CXCR-4 signaling cascade.Based on our findings, hesperidin maybe a promising drug against NSCLC.Cell migration is a normal physiological process, required for chemotaxis of white blood cells and directional movement of cells from other tissues[27, 28] . However, in tumor tissue, the metastatic ability of malignant tumor cells is significantly associated with the likelihood of patient death[29] . Previous studies have shown that the activation of tumor cell migration is associated with the activity of the SDF- 1/CXCR-4 signaling pathway[30-32] . The SDF- 1-activated CXCR-4 receptor, which is folded and delivered to the cell surface by HSP90, induces a phosphorylation cascade, leading to actin reorganization and subsequent cell migration[33] . In the present study, we report that the SDF- 1α-mediated enhancement of migratory ability in A549 cells could be inhibited by treatment with the SDF- 1/CXCR-4 specific inhibitor AMD3100 or hesperidin. Based on the results of ELISA and western blotting, we confirmed that hesperidin could significantly reduce the protein expression of CXCR-4 and the level of SDF- 1 secreted from A549 cells. Thus, hesperidin may potentially inhibit tumor cell metastasis through a complicated process. Interestingly, a recent study by Li et al. PF-06882961 order found that the natural product Kongensin A was a non-canonical HSP90 inhibitor that blocked RIP3-dependent necroptosis, providing a new perspective on the matter[34] . This informed our hypothesis that the inhibitory effect of hesperidin on the migratory and invasive capabilities of human NSCLC cells may be mediated by the regulation of SDF- 1 and downstream molecule expression and activity, through target proteins such as HSP90, although further study is required prior to the development of hesperidin as a therapeutic drug.

In the present study, we identified that SDF- 1α could significantly elevate the expression of p-P65 in the NF-κB signaling pathway and p-Akt in the PI3K-Akt signaling pathway, while significantly reducing the expression of p-IκB in the NF-κB signaling pathway. AMD3100 and hesperidin antagonized the effect induced by SDF- 1α, suggesting that hesperidin inhibited the migration of A549 cells by blocking the SDF- 1/CXCR-4 signaling pathway. Furthermore, the present results demonstrated that hesperidin promoted the activation of the NF-κB signaling pathway. However, our results indicated that hesperidin could increase the level of p-IκB, which is contrary to previous studies in human hepatocellular carcinoma cells and Ramos cells[35, 36], suggesting different effects of hesperidin treatment in different tumor types. In addition, the results demonstrated that hesperidin inhibited the migration and invasion of A549 NSCLC cells in a time- and dose-dependent manner, as demonstrated by wound-healing and Transwell assays, which provides a theoretical basis for further trials on NSCLC cell metastasis in rat models.

EMT, the process of cohesive epithelial cells converting to a migratory mesenchymal phenotype, is regarded as a contributing factor in the metastasis of tumor cells[7, 37] . Our results indicated that the expression of CK- 19, a cytoskeletal protein specifically expressed in epithelial cells, was evidently upregulated in hesperidin-treated A549 cells. Similarly, the protein expression of Vimentin, a mesenchymal marker, and of MMP-9, a mesenchyme-associated molecular marker that serves a vital role in digesting the extracellular matrix to promote tumor cell metastasis[8][38, 39], was downregulated compared with the control group. This indicated that hesperidin may prevent tumor metastasis by inhibiting the EMT phenotype transformation. In addition, EMT-related proteins were previously reported to be the regulatory targets for downstream molecules of the SDF- 1/CXCR-4 axis[40] .Therefore, the inhibition of EMT by hesperidin is of great interest for the treatment of tumors.

Taken together, our results demonstrate that hesperidin inhibited the migratory and invasive capabilities of A549 NSCLC cells, which may have been mediated through blockade of the SDF- 1/CXCR-4 pathway. We also confirmed that hesperidin inhibited the migratory ability of H460 and H1975 cells via a wound-healing assay, reflecting the potential clinical value of the data from A549 cells. Further studies will be required to fully understand the underlying mechanisms of hesperidin in NSCLC and lay the foundation for the application of hesperidin in the clinic.
